The bacterium staphylococcus aures was isolated from subcutaneous abcess by using staphylococcal medium No-110 and diagnosed according to its cultural and micro scopic characters , biochemical reactions and coagulase production . Sensitivity test of the bacterium against antimicrobial agents was performed and the result appeared that the bacterium was sensitive to cephlothin , ampicillin , ampiclox , tetracyclin , rifampicin , cloxamcillin ,fusidic acid , chloramphenicol and nitrofurontion , and resistant to cefotaxin , bacitracin neomycin, gentamycin , nalidixic aceid , penicllin , erythromycin and cloxacillin . Curing experiment was accom plished using elevated growth temperature , to study the role of plasmids in coding for the virulence factors , coagulase and hemolysin and antibiotics resis tance , Result showed the partial loss of coogulase production through the effect on RNA polymerase responsible of transcription of cluster structural genes with single promoter and terminator produce coagulase and the method was not suitable for curing hemolysin plasmid . Results of curing of plasmids responsible of antibiotics resistance ensured the role of plasmids in resistance of the antibiotics penicillin , Erythromycin and cloxacillin after physical loss of plasmid as proofed by gel electrophoresis .
