Read More ... Forty specimens of surgically removed hydatid cyst were obtained from patients in different hospitals in Al-Anbar province. The aim of study was designed to determine the type of common larval stage (hydatid cyst) of echinococosis in Al-Anbar province according to PCR molecular differentiation.The study was carried out during the period from first day of January to the last day of December 2013.The age of patient ranged from (10 – ≥ 65 ) years old, (19 males and 21 females). A questionnaire paper was performed to each patient which includes information about age, sex,residence,occupation,exposure variable, water source, site of hydatid cyst and case history.From the obtained results, it's revealed that the age group (21 – 30 years) presented high hydatidosis parasitic infestation which was 35% , whereas the age group more than50 years presented low parasitic infection which was 2.5% . 47.5% of patients were males and 52.5% were females.Twenty eight(70%) patients live in rural areas. Also, twenty patients (50%) were exposured to livestock animals, (40%) of patients were farmers, twenty seven patients(68%) were consumed river water, The most common localization was the liver (65%). Purification of DNA was done by QIAamp DNA Mini kit from QIAGEN for cyst. Specific primers were designed for the diagnostic PCR. It amplifies a 133-basepair segment within the newly described repeated unit that amplified a target repeated sequence (EgG1 Hae III) newly identified in the genome of the common sheep strain of E. granulosus. Primer based on the flanking sequences allowed the PCR amplification of E. multilocularis, a fragment of mitochondrial 12S rDNA was amplified by PCR from human genomic DNA by using the cestode-specific primers 60 were used as targets for (PCR) amplification.The total number of the cysts specimens that gave positive results of E.granulosus by PCR was 37 (92.5%) patients and the number of specimens that gave negative results was 3(7.5%),while all specimens gave negative result of E. multilocularis by using PCR analysis.The present study showed that E. granulosus is a major causative of hydatidosis in Al-Anbar Province while E. multilocularis is not detected by using PCR approach.

Pneumocystis jirovecii is an opportunistic eukaryotic pathogen causing life threating pneumonia ( PJP) in immunosuppressed patients.Detection of Pneumocystis jirovecii Pneumonia in immune suppressed patients like hematological cancer with or without chemotherapy, patients on corticosteroid therapy, other malignancy like bronchogenic cancer by using External Polymerase Chain Reaction (External PCR) and to compare the results with other control group (patients who are immunocompetent suffer from pneumonia).A total of 220 specimens were obtained from patients admitted to Al- Ramadi Teaching Hospital, Clinic excitability of respiratory disease in Al- Ramadi and Oncology Teaching Hospital in Baghdad during the period from February to July 2013. This number composed of 71(32%) specimens were from female while the male specimens were 149(68%). A total of 184 (84%) were immunocompromised patients, They were considered immunocompromised patients because they have ( hematological cancer with or without chemotherapy, patients on corticosteroid therapy, other malignancies like bronchogenic cancer). The other 36 (16%) patients with pneumonia were included as immunocompetent patients (No hematological cancer, No chemotherapy, No immunosuppressive drug like corticosteroid therapy and No malignancy like bronchogenic cancer). A total of 220 specimens which were collected, (consisting of 115 blood, 65 sputum, 40 BAL Specimens )the total 220 specimens (115 blood, 65 sputum and 40 BAL Specimens ) were examined PCR technique ( External and Nested PCR ) to identify Pneumocystis Jirovecii DNA based on the amplification of specific primers. The result of agarose gel electrophoresis revealed that small DNA diagnostic bands were detected at 346 bp for External PCR in all positive specimens. Out of 220 specimens 26 (11.8%) were detected by the mtLSU rRNA External PCR. Regarding the use of External PCR technique for sputum specimens 8 (12.3%) were positive, While BAL 8 (20%) and Blood 10 (8.7%) . PCR is more accurate and better indicative for the presence of Pneumocystis jirovecii Pneumonia. In addition this technique can use specimens which were not stored properly and became non suitable for microscopy.

This research included the estimation the concentration hemoglobin and the levels of some its derivatives which include the Oxyhaemoglobin, Methamoglobin, Carboxyhaemoglobin and Sulfhaemoglobin in the blood of women during delivery (90 sample) and compared it with 90 samples of their levels in the umbilical cord blood of their newborns (43 female and 47 male). The samples of this research were collected from the newcomer women to the general hospitals of Al-Fallujah and Al-Ameria and their newborns ,women ranged in age from 18-40 years old and who are apparently healthy, and included both natural (vaginal) and caesarean delivery. The findings of this research reached to find high significantly differences (P≤0.01) in haemoglobin levels for the group of women during delivery and newborn with average (11.676 ± 1.0) and (16.566 ± 1.8) g/dl at the succession.Also it finds high significantly diferences (P≥0.01) between the levels of Hemoglobin in women blood during the delivery compared with those of their newborns. Their levels reached (0.611 ± 0.1 , 0.604 ± 0.1 , 0.557 ± 0.9 , 0.530 ± 0.9) for each derivative of Oxyhaemoglobin, Methmoglobin, Carboxyhaemoglobin and Sulfhaemoglobin at the succession in women blood during the childbirth. While their levels reached to (0.783 ± 0.13 , 0..768 ± 0.13 , 0.721 ± 0.11 , 0.677 ± 0.12) in the umbilical cord blood of their newborn for the same derivatives at the succession.It also found a simple positive correlation (r = 0.259) between each of the concentration of the haemoglobin of the mother and haemoglobin of their newborns and found that the weight of the mother influence the values of rates of the derivatives Carboxyhaemoglobin and Methmoglobin for the newborns, as values were significantly higher (P=0.05) in category gravimetric (60-50) kg , compared with other categories of these derivatives, while the derivative of the Oxyhaemoglobin arose significantly(P=0.05) in the age group (80 and over) kg. While the haemoglobin and its derivatives not affected both in the blood of newborns with the age of mother , type of delivery, and sex , weight of the newborns.

Read More ... الوراثة اللاجینیة تعنی التغیرات والأنماط المظهریة التی تظهر على الکائن دون المساس بتوالیات مواده الوراثیة DNA sequences . وقد أعطیت أکثر من تعریف ولکن کلها تصب فی هذا الاتجاه وقد سماها البعض الوراثة الجدیدة New Genetics . وبعض هذه التغیرات یمکن ان تتوارث ، فضلا عن ان اغلبها قابل للرجوع (2،1) والدراسات فی هذا المجال تشکل ما یسمى بدراسة الجینوم اللاجینی Epigenomics (3) وتعنی دراسة الجینوم المبرمج Programmed Genome . فالملاحظ ان الخلایا حقیقیة النواة تستنسخ معظم DNA المکون لجینومها ولکن ما یستعمل منه للتشفیر للبروتینات یقع بین 1-2% . وبالرغم من ان کل خلایا الکائن الحی مثل الإنسان تحوی المادة الوراثیة نفسها ، الا انها تختلف فی الشکل والوظیفة التی تؤدیها ، وتفسیر ذلک یکون بواسطة التنظیم اللاجینی Epigenetic Regulation للجینات وبمثل هذا التنظیم یحدد من الجینات وکمیة نواتجها ویتم تجاوز الجینات الأخرى ( 4) ، وهذا ما یمکن ان یفسر ان التوائم المتماثلة الحاویة على جینات متماثلة یمکن ان تظهر علیها اختلافات فی الشخصیة واختلاف فی الأعراض التی تصاب بها ( 5) . • فی السابق استعملت الوراثة اللاجینیة لوصف عملیات تمایز (Differentiation) الخلایا المتماثلة من الناحیة الوراثیة التی تؤدی الى انتاج أنسجة وأعضاء خاصة أثناء نمو الأحیاء متعددة الخلایا ( 6) ، اما فی الوقت الحاضر فهی تستعمل من قبل العاملین فی مجال علوم الحیاة لوصف التغیرات المؤثرة وربما المستورثة فی التعبیر الجینی دون مساس توالیات DNA (6) ، وهذا یعنی ان الواسمات اللاجینیة یمکن ان تعبر للأجیال القادمة نظرا لعدم محوها بالکامل فی المرحلة التی تعقب تکون البیضة المخصبة Zygote بعد اتحاد الأمشاج Gametes وهذا ما یعقد التوارث المندلی المتعارف علیه ( 1) . وفی السنوات الأخیرة ظهر حقلین من الدراسة هما • الوراثة اللاجینیة التی تعنی بدراسة Epigenome • الکیمیاء الحیویة للکروماتین Chromatin Biochemistry ان فکرة الوراثة اللاجینیة لیست بالجدیدة فهی وصفت فی عشرینات القرن المنصرم عند اکتشاف طفرات ثابتة فی ذبابة الفاکهة Drosophila melanogaster ونبات الذرة Zea mays . وهذه الظواهر أدت بشکل جاد الى ظهور کیمیاء الکروماتین بعد عدة عقود ، وبرزت بشکل کبیر فی سبعینات القرن الماضی عند اکتشاف ان المواد الوراثیة مع البروتینات الملحقة بها مثل الهستونات تنظم فی تراکیب خاصة هی الجسیمات النوویة Nucleosomes وأدت الى التعرف على العدید من البروتینات غیر الهستونیة وجزیئات أخرى تکون مسئولة عن تنظیم الترکیب الثلاثی او التوزیعی للکروماتین ومسئولة عن معالجة الکروموسومات داخل الخلایا والتی تشمل عملیات
